A homicide detective said it was unlikely missing toddler Daniel Thomas could have left his Myrtleford home in north-east Victoria alone.

The homicide squad has taken over the probe as a murder investigation after it was revealed two-year-old Daniel was left alone for three hours before he was reported missing last Friday.

Detective Senior Sergeant Rowland Legg said the homicide squad had been involved in the investigation since Saturday but had now taken a higher level of interest.

"We've established a number of things, one of which is we now believe it's quite unlikely the child would have been able to leave the house of his own accord," Det Sgt Legg told Channel Nine.

Daniel was being looked after by a family friend when he was reported missing about 4pm (AEST).

"The information we have at the moment ... is that he was left in the house alone for approximately three hours in that afternoon," Det Sgt Legg said.

About 80 police and State Emergency Service volunteers combed Myrtleford, widening the search to include underground drains, dams and the Ovens River.

A police spokeswoman said search would continue.

Several people have been interviewed over the disappearance, including neighbours and trades people working in the area.

Daniel suffers asthma and wears heavy bandages due to severe eczema, making walking difficult.

Oh dear this is so very sad. Another young soul missing. Poor Daniel, I shall pray for him. This brings back memories or Coral Burrows. I hope he will be allright.

this is so very good poem. it's sad and true i hope they find him and he's okay. i don't understand how can people do that. i mean i ruin a daycare and i love the kids. i mean they may get to me but i still love them. i just don't understand how can someone bring them into the world and hurt them. i love them so dear and they not even mind.
